Your Body Could Be Immune to Gene Editing, What Now? - Duration: 3:16.

A lot of hay has been made about CRISPR Cas9, the gene editing technique so precise, so

easy, so versatile it's opened the possibility of chopping and swapping DNA around like a

molecular level Dr. Frankenstein.

The implications are huge for humans and our genome.

We could alter the genes of embryos, or treat genetic maladies in those already born.

Now in 2018 clinical trials on humans are scheduled to start for the first time.

But just as they're getting underway, there may be a problem thanks to our immune system.

To understand why our immune system is being a jerk and preventing us from treating disorders

like sickle-cell anemia, it helps to step back and remember exactly what CRISPR Cas9

is.

It's actually an immune response bacteria use to protect themselves from viruses.

Cas9 is a protein the bacteria make that act as molecular scissors, cutting virus DNA to

ribbons, and two of the most commonly used types of Cas9 come from bacteria that can

cause staph infections or strep throat.

These are bacteria we want our immune systems to have a defense against.

And to defend against them, our immune system has to be able to recognize the bacteria and

the various proteins they make, so they can track them down and destroy them.

Sorry I called you a jerk earlier, immune system, you're just doing what comes naturally.

To see if these two versions of Cas9 were on our immune system's hit list, researchers

tested the blood of 34 donors for antibodies.

They found 65 percent had a defense against the strep version while 79 percent fought

the staph variety.

In another experiment with 13 blood donors, 6 of them had t-cells that attacked staph's

Cas9.

The findings are yet to be peer reviewed, but one of the authors is the scientific founder

of a leading CRISPR human therapy company, so he's got some credibility.

And if our immune systems respond to CRISPR, it could mean more that just rendering the

therapy ineffective.

It could make our immune systems go haywire and kill us, which has happened in a previous

attempt at human genetic therapy.

The next question is: is this the end of the line for CRISPR?

Good game, well played, but our bodies are just too vigilant for this revolutionary gene

editing?

Hahaha, no, we're going to science our way around this conundrum!

There are already a few solutions to this possible problem, like modifying Cas9 into

a form our immune system can't recognize, or using a different protein from the oodles

of other bacteria that have their own versions of Cas9.

Or if the protein gets attacked when it's in our bodies, then perhaps we could put it

to work outside our bodies, modifying extracted cells before returning them to the body.

Or Cas9 could be used in places where the immune system is locked out, like inside our

eyes where it could treat hereditary blindness.

CRISPR has way too much potential to let a little thing like our bodies stop progress.

The findings of these recent experiments are more of a speed bump than a brick wall.

Medical advances occasionally have to overcome biological defenses.

For example our blood vessels in our brains make it really hard to deliver medicine to

our old noodle.

IT'S HAPPENING NOW!!!

The world's largest active geyser has erupted three times in the past six weeks at Yellowstone

National Park, including once this week, in a pattern that is highly unusual, according

to geologists.

And scientists cannot explain what is causing the geyser to erupt.

One thing all scientists can agree on is that, as they've repeatedly told us before, the

supervolcano churning below the serene scenery in Wyoming is not going to erupt anytime soon.

We shouldn't be taking any of the anomalies to mean as much either.According to Reuters,

the Steamboat Geyser, which can shoot water as high as 300 feet (91 meters) into the air,

erupted on March 15, April 19 and on Friday.

The last time it erupted three times in a year was in 2003, the U.S. Geological Survey's

Yellowstone Volcano Observatory said.

The last time it erupted prior to March was more than three years ago in September 2014.

As always, there's nothing to worry about.

"There is nothing to indicate that any sort of volcanic eruption is imminent," Michael

Poland, the scientist in charge for the observatory, said in an email.

This year's eruptions have been smaller than a usual Steamboat eruption, but the two

in April were about 10 times larger than an eruption at the park's famed Old Faithful

Geyser in terms on the amount of water discharged, he said.

Geologists have not pinpointed a reason for the latest series of eruptions, but say they

could indicate a thermal disturbance in the geyser basin, or that Steamboat may be having

smaller eruptions instead of one large.

–Since most geysers do not erupt on a regular schedule, "it might just reflect the randomness

of geysers," Poland said.

While the Steamboat eruptions are unusual, what would be far more worrying would be the

water in the hydrothermal systems drying up, which could indicate that the super hot magma

deep below was making its way to the surface -and an eruption could occur.

So here it is, when you get close to the little asteroid, you'll see a small crashed boat

and that's exactly where the location will be.

INVOLUTION INNER TRANSFORMATION AND SELF-AWARENESS

There�s a mess inside you: You clean the outside. ~ The Dhammapada

In life we�re confronted with many puzzling, cryptic and complex questions.

Who am I? Why do I keep making the same mistakes? Why do I continue suffering so much pain?

What am I supposed to be doing with my life? Where am I going?

In our slow and tumultuous journeys we come across hundreds of possible answers, and thousands

more dogmas, creeds and doctrines that claim to cure our inner sicknesses and give meaning

and direction to our confusing lives.

At the end of the day, many of us forget a very simple truth:

Change � deep, long-lasting change � comes from within.

Yet many of us outsource and externalize our efforts in an attempt to change the way we

think, behave and interact with ourselves, other people and the world.

This is where Involution comes in.

Involution, the antithesis of evolution, does not align itself with any particular dogma,

creed or religious doctrine, but is rather an experiential practice of life.

The practice of Involution is an all-encompassing philosophy; one that can be observed in the

actions of thousands. From the Sadhus, yogis and monks of the East, to the New Ageists,

fitness nuts and self-help enthusiasts of the West, Involution can be observed anywhere

in which a person or group of people seek inner strength, understanding, metamorphosis

and liberation.

SELF-AWARENESS AS A TOOL OF INNER EVOLUTION Without awareness, we are not truly alive.

Are you a self-aware person? Think about this question for a minute.

How conscious are you really of the many thoughts, feelings and behaviors you exhibit during

the day? If you have a tendency to:

Be unaware that your words or actions have hurt or offended another person,

Be self-destructive and continue to make the same mistake over and over again,

Have trouble identifying what exactly you are feeling or thinking (poor emotional intelligence),

Have a habit of behaving without thinking, Have an unclear perception of who you (think)

you are, � chances are that you are low on the self-awareness

spectrum.

DEVELOPING SELF-AWARENESS The most fundamental aggression to ourselves,

the most fundamental harm we can do to ourselves, is to remain ignorant by not having the courage

and the respect to look at ourselves honestly and gently. ~ Pema Ch�dr�n

Self-awareness is the essential building block of inner transformation because without being

aware of who we are and what we do, there is little chance that we can grow to understand

why we feel, think and behave the way we do, and thus grow closer to living a life of peace.

To become a self-aware person requires patience and practice. There are no quick fixes. There

are no immediate solutions. In order to follow the path of Involution, we must be persistent

in our efforts. These efforts could include:

Paying Attention. Consider for instance: what you are saying,

how this is influencing the people around you, what you are feeling, how this is influencing

your behavior, what you are thinking, when your thoughts occurred, what you like and

dislike, what makes you happy and what makes you sad. Paying attention to what goes on

inside of you is an excellent way of developing self-awareness.

Mindfulness Meditation. There�s no need to practice this for even

10 minutes a day. 5 minutes at first will suffice, but in order to make that 5 minutes

count, you must make a habit of this practice. Start ideally first thing in the morning,

in a quiet, comfortable place. Mindfulness meditation is the practice of becoming aware

of your inner chatter, in the present moment and letting it pass. Benefits include increased

inner calm and stability, and decreased emotional reactivity.

THE BOAT ON TOP OF THE OCEAN Until you make the unconscious conscious,

it will direct your life and you will call it fate. ~ C. Jung

The non self-aware person can be compared to a little boat drifting on the surface of

the ocean. The stability of that boat is completely reliant on the stability of the ocean beneath

it. When the sea is turbulent and chaotic, so too is the boat; cast here and there, to

and fro violently.

The same can be said of the non self-aware person; their peace and happiness is entirely

controlled by how calm and stable their inner worlds are. In fact, the non self-aware person

is basically a slave of their thoughts and emotions, which is the main culprit for their

suffering.

The intensity of the person�s servitude is dependent on the intensity of their inner

turmoil. This is the principle of the Boat On Top Of The Ocean.

FREEDOM FROM SLAVERY Never become a slave to your own thinking

and your own self. Understand that you are continuously changing. But, in which way you

will change is your free will. ~ R. Papian

It may strike you as an odd thought to think that you enslave yourself to yourself. But

when we lack self-awareness, we perpetuate the pain and suffering in our lives, both

within and without ourselves.

Involution can help address these problems. When we follow the process of Involution,

we realize that self-awareness is an essential building block of inner transformation.

some people work for a travel agency or

they have different skills

Right now, we just started so we just have monthly meetings

and just like introduce ourselves and then catch up

I haven't met everyone in this organization

So like every time it's kind of like you know, a nice to meet you kind of thing

but it's really nice to know like what other people are doing

So it's a good opportunity to expand your network

So right now, I don't really know what's going on like nobody knows what's going on

because we just started, but it's gonna be very exciting so yeah

hey guys today I'm going to show you how to use our WODFitters lifting belt to

provide some extra support and your lower back for your hips alright we're

going to take the velcro strap opening it up you're gonna clip it through the

side clip crossing over and making sure it's nice and tight right above your hip

bones so three lifts that you can use your belt for are dead lifts you can

move from there into a front squat

and last but not least you can use Romanian deadlifts as another way to

support your lower back with your weight training belt on
