Cheesy Ground Beef & Potato Casserole ~ Hamburger Potatoes Recipe - Duration: 4:46.

Hi everyone I'm Tess and tonight I'm making a hearty comforting and

budget-friendly dinner. Come join me in my kitchen while I make

an easy beef and potato casserole.

This cheesy ground beef and potato casserole

is going to hit the spot when on this very cold evening.

I love making casseroles because they are easy, comforting, budget-friendly and

feed a large family. This casserole will feed up to 6 hungry people. In a skillet on

medium heat I have about a pound and a half of ground beef. Cooking and chopping

into small pieces for about five minutes or until no longer pink. I'm going to

drain some of the fat because I don't want my casserole greasy. I'm bulking up

the casserole and adding in some flavor with some vegetables. You can season the

ground beef and add just the meat but I like adding the veggies and you can add

what you like or have. Adding in some finely diced onions, bell peppers, carrots

and a little salt and black pepper. Cooking and stirring for a couple

minutes and I'm also adding in some minced garlic and I also found some

frozen peas that I need to use. Giving that a stir for a minute and then I'm

turning off my heat. The vegetables may not be tender but they will finish

cooking in the oven. For the sauce I have two cans of cream of mushroom soup.

You can use cream of chicken or celery. Adding in a little salt and not much.

Black pepper, garlic powder, onion powder, about a cup of milk, Worcestershire sauce

and about 1/2 a cup of sour cream. You can add the seasonings that you like.

Giving that a good mix.

I have my oven preheating to 350 degrees Fahrenheit. In a well-buttered or greased

9 by 13 pan I'm layering the bottom with some sliced yellow potatoes. The slices

are about 1/8 of an inch thick and I'm just overlaying them a little to cover

the bottom. I have a little of the onion left over so I'm evenly distributing that

on top of the potatoes. Next on goes the meat and vegetable mixture. I split the

mixture in half and I'm sprinkling evenly over the top. Half of the liquid

gets poured over the top and just spread out as evenly as you can.

Half of the grated cheese goes on next and I have a mixture of Colby jack and

cheddar... and don't be chintzy with the cheese.

And now we're just going to repeat the layers; a layer potatoes, the meat and

veggie mixture and finally topped with the remainder of the sauce and spread

out evenly. This gets covered and baked at 350 degrees

Fahrenheit for one hour and then I'll bring you back.

It's been one hour and the kitchen is smelling amazing. I'm taking the foil off

and topping with a remainder of the cheese. This goes back in the oven for

another 30 to 40 minutes or until the potatoes are tender and the top is

cheesy bubbly golden goodness.

The cheesy beef and potato casserole is done and piping hot. Going to let this

cool for about 20 minutes before cutting. As you can see this will feed up to six people.

With a side salad and a piece of bread this is a comforting meal.

The potatoes are nice and tender and I love the rich beef flavor. The veggies add a

lot of bulk, flavor and texture and the cheesy sauce just brings it all together.

Your family and pocketbook will love it. I hope you give this cheesy ground beef

Wear Multiple Watches At The Same Time? Can A Man Wear 2 (Or More) Watches & Be Stylish? - Duration: 7:14.

In today's video, gents, how to wear multiple watches at the same time.

[Music] So, I got this question years ago, Antonio,

can a man wear more than one watch at the same time and still be stylish?

At the time, I totally discounted the question like I'm not going to make a video of this.

But, I got this question again and, now, that I love watches and I've got actually some,

I have found myself wearing more than one watch at the same time.

So, I decided to create this video to actually make the case that a man can wear more than

one watch at the same time and still be stylish.

So, let start this video off by answering the question why.

What are reasons that men would want to wear more than one watch?

Okay.

So, imagine you're the CEO of a company and you've got a status watch.

The original James Bond Rolex Explorer because you explore the world and this is a symbol

of what you've earned.

But, right below that you want to wear your old trustee Databank.

Why?

Because you started off as an accountant.

You love to calculate numbers and you want a calculator handy, but you also go diving

in the afternoon to take a private jet down the Bahamas and so, you've got your trustee

diving watch.

Now, the point I'm making with that quick example is that different watches serve different

purposes.

One watch you may wear because of status because of just the design.

You love the way it looks.

Another one, because of the function because of what it reminds you where you came from.

Another one, because it can go down to 200 meters, it's got different time zones, you've

got people working in five different areas.

I know for me, I've got people that work for me in nine different time zones.

So, guys, understand that you can actually bring in different watches and if you treat

it like jewelry, maybe wear one on each wrist.

I'm going to say that you're going to wear them all on the same wrist, but what

you're looking here is you're wearing different watches that serve you in a different

way.

Again, two different watches, two different purposes, both of them very durable, but that's

where the similarities really end between these watches.

This one got set with atomic time.

This one I can actually get the temperature, I can measure my heartbeat.

This one is a very smart device.

I can go through the mud with this one.

You probably don't want to go with the mud with this one, but if I'm going to be on

a yacht, hey, wear the Yacht Master.

The point is different watches different purposes, they serve different functions.

Another reason to wear two watches, you simply like the look.

You want to be different, you want to stand out from the crowd.

You want to get questions about this.

You like the way that this white dial makes the blue dial look even bluer.

The simply the contrast, the silver works together, but here you've got the leather

strap with the metal strap.

You like the contrast.

Again, you do it for yourself.

Maybe for some of you guys, you feel it brings balance to your outfit.

You know you just feel weird with the watch on one arm, so why not put it on both that

way you balance it out.

And I've heard some people even cite safety concerns especially when they depend on their

watch.

Let's say you're a deep sea diver and you need to know the time, you need to be

able to split time, you need to be able to use this information to make important decisions.

You're a first responder and you don't want to rely on just one watch where the battery

could go dead or the other one it just get smashed and gets destroyed, you want to have

a back up.

In that case, why not wear two watches.

And a quick bit of related trivia when Sir Edmund Hillary first climbed Mt. Everest,

he took two watches with him.

Why?

Because you got to be able to measure how much oxygen is left in the tanks.

They didn't have a measuring device, so they used timers.

Which watches did he take?

Guys, one of them was a Rolex, the other one was which brand?

The next reason wearing more than one watch at the same time is that you're going to

get more wears out of those watches.

So, let's face it, your collection has grown, you've got a modest collection of fifty

watches which means if you wear one watch per day and you rotate through, seven wears

per year.

Guys, that's not enough.

You love your watches, you want to wear them show them off more, so you start to wear more

than one at the same time and guess what?

You're able to wear them more throughout the year and kind of pass the love around.

Finally, and this one is straight and practical.

When I'm traveling I don't want to leave my nice watch in my luggage, so guess what?

I'm wearing two watches through security that way I can keep track of them.

I know where they're at I'm not going to damage them and I just don't want to

lose my luggage and then lose one of my nice watches.

So, I end up wearing multiple watches when I'm traveling if I'm taking multiple watches

with me.

So now, let's talk about the dangers of wearing multiple watches.

The first one is that you look too awesome and you're just going to blow people away.

No, seriously, people are going to ask questions, they're going to look at this, they're

going to like, ah, what's going on here?

And, yes, this is overkill.

But what I want to stress here also is you've got watches rubbing again each other, so you

will get scratches on your watches if you wear multiple watches.

Another thing, if you've got watches that are basically mechanical and they've got

very intricate movements in there, you got to be careful because knocking up against

other watches can actually damage the inner mechanics.

So, how to pull off the look?

Well, you can go on each arm, the balance look.

Wearing an odd number of watches than one side you can make the strong side.

Now, you can't wear both watches on one arm, understand this one is going to get stronger.

And if you're going to do this, I like to go for a little bit of contrast.

And, finally, pay attention to proportion.

Smaller watches should be closer to the wrist, the larger watches can make their way down

the forearm.

Video gambling: Not a great way to fund a government - Duration: 5:59.

This is a video gambling machine.

Across the U.S., you'll find more than a 100,000 of them offering up electronic versions

of games like poker or slots.

The machines — designed to be highly entertaining and addictive — have taken the gambling

industry by storm.

23 states have them in state-sponsored casinos or other gambling establishments, and now,

more states are considering legalizing them outside casinos — in places like bars and

restaurants.

But, why?

We looked at the state with the most locations to legally place a bet in America.

No, not Nevada.

Illinois.

People love gambling.

This has been true for centuries...across cultures.

And in the US — it's also been a source of government revenue.

When the colonies needed to build hundreds of schools in the 18th century — they got

funding from some of America's first lotteries.

The Washington Monument?

Also funded in part by lotteries.

And when the Great Depression hit, Nevada legalized gambling.

States across the country continue to count on it to bail them out of tough financial situations.

Take Illinois.

In 2009, the state passed the Video Gaming Act, its largest gambling expansion since

the creation of the state lottery in the '70s.

The law paved the way for video gambling machines to go into truck stops, restaurants — even

hair salons.

Today, more than 30,000 of these machines operate outside of casinos in Illinois — more

than any other state in the country.

The problem is — the machines can be highly addictive.

The algorithm-driven machines are essentially made to do three things: accelerate play,

extend the time people play them, and increase the amount of money they wager.

For example — on old-fashioned slots, you pulled a lever that spun physical reels.

Each reel had a certain number of symbols and blank spaces on them, and depending on

where they landed, if the symbols lined up across this payline, you'd win a jackpot.

But with electronic machines — while you may

see virtual reels spinning, the symbol that the reels land on is no longer determined by the

pull of a lever. It's decided by an algorithm the moment you click play.

The reels can be programmed to have more losing symbols and fewer winning ones, so game designers

can reduce the odds of hitting a jackpot.

They could make a machine with over 500 stops on the reels,

meaning the odds of winning would be less than

1 in 100 million.

The virtual display also creates deceptive tricks like the "near miss", when jackpot

symbols appear right above or below the winning line — it's an image that can make

players think they were so close to winning the jackpot and it's designed to keep them going.

These characteristics have earned the machines nicknames like "electronic morphine" and

"the crack cocaine of gambling."

About a decade ago, Illinois made these machines widely accessible across

the state.

"The economy still reeling from a recession."

"This is a financial crisis-led recession."

"The worst recession since the 1930s."

During the financial crisis in 2009, Illinois wanted money to help fund a big infrastructure

program that would create jobs.

"We know our economy needs a boost, our economy in our country, our economy in our state."

So, tucked inside a larger bill, the state passed the Video Gaming Act.

They agreed to expand gambling in the hope that the revenue from video gaming taxes would

help pay off their debts.

The state started borrowing money, assuming it could pay much of it back through taxes

from video gambling.

But here's the thing: It took more than three years to get video gambling up and running.

By the time video gambling went live, in September 2012, the state had already borrowed more

than $5 billion.

Debt payments had reached about $340 million

a year.

While the machines were supposed to raise $300 million a year to cover those debt payments,

in 2012, video gambling brought in just $31 million.

A decade later, the industry has finally started hitting revenue projections.

But the Illinois Gaming Board,  the agency tasked with keeping gambling in check, is

largely underfunded and has struggled to regulate the growing industry.

A small group of gambling companies has made nearly 2 billion dollars.

And politicians have received hundreds of thousands of dollars in contributions from

an industry group.

Meanwhile, in rural areas and small towns hit hardest by the state's economic slump,

gambling addiction has become a serious problem.

But the state now spends less on addiction services than it did before legalizing video

poker and slots.

What's happened in Illinois could be a sign of what's to come in the rest of the country.

A 2018 Supreme Court decision legalized sports betting, paving the way for a new frontier

for gambling in the US.

As more states look to gambling as a possible

way to stabilize their finances, Illinois' experience with video gambling stands as a

cautionary tale.

That maybe, it's a risky bet.

AMERICAN B2 SPIRIT STEALTH BOMBER NOW WITHIN STRIKING DISTANCE OF CHINA - Duration: 5:55.

Pacific Air Forces Public Affairs has revealed, the US has deployed three B-2 Spirit bombers

and 200 airmen to Joint Base Pearl Harbor-Hickam in Hawaii for training in the Pacific.

The B2s were deployed to the Hawaii from Whiteman Air Force Base and are intended to support

US Strategic Command's Bomber Task Force mission.

US Strategic Command's Bomber Task Force mission is a deterrence undertaking aimed at maintaining

regional peace and reassure allies.

Lt. Col. Joshua Dorr, the director of operations for the 393rd Bomber Squadron stated,

"Deploying to Hawaii enables us to showcase to a large American and international audience

that the B-2 is on watch 24 hours a day, seven days a week ready to protect our country and

its allies.

This training is crucial to maintaining our regional interoperability.

It affords us the opportunity to work with our allies in joint exercises and validates

our always-ready global strike capability."

The latest deployment marks the second time B-2 Spirit bombers have been deployed to Hawaii.

The deployment comes at a time when there is increased tension between U.S & China and

clearly indicates America's willingness to stand up to China.

B2 Spirit brings a whole new level of capability to the table.

In this video Defense Updates, analyzes why China should fear B2 Spirit bomber deployment in Hawaii?

Let's get started.

The B-2 Spirit was developed to take over the United States Air Force's (USAF) vital

penetration missions, able to travel deep into enemy territory to deploy their ordnance,

which could include nuclear weapons.

The B-2 is a flying wing aircraft, meaning it has no fuselage or tail.

The blending of low-observable technologies with high aerodynamic efficiency and large

payload makes B 2 Spirit the most potent bomber in the world.

Even after around two decades from its introduction on 1 January 1997, there is still nothing

to match it.

America's rivals Russia and China have so far not been able to field a proper stealth

fighter let alone a stealth bomber.

Chinese have no radar that can detect or track it.

B-2 Spirit has a maximum speed of Mach 0.95.

The U.S. Air Force reports B2's range as approximately 6,900 miles or 11,000 km.

With this range, it can reach mainland China if required.

B2 has an excellent payload, which makes even one of these bombers to be extremely destructive.

It has 2 internal bays for ordnance and payload with an official limit of 40,000 lb (18,000

kg) and maximum estimated limit of 50,000 lb (23,000 kg).

Apart from different types of conventional bombs, B 2 can carry 16× B61 or B83 nuclear

bombs on Rotary Launcher Assembly (RLA).

The B61 nuclear bomb is the primary thermonuclear gravity bomb in the U.S. cache.

How To Say Common English Expressions! 💬 Small Talk PART 2 - Duration: 15:22.

Well hey there! I'm Emma from mmmEnglish!

In this lesson, we'll focus on the natural pronunciation

of some very common English expressions.

We'll study how a native speaker would

really use these expressions and practise saying them

together, exactly as a native speaker would.

Common expressions, ones that are used all the time,

are not often pronounced as you would expect.

As native speakers, we say them quickly,

the sounds reduce, they even disappear.

And even simple expressions become

difficult to recognise if you're not used to hearing them.

In my lessons, you hear me speak really clearly

and a little slower than I normally would

and while that makes you feel good like,

'Yes! I can understand everything she's saying.'

you do need to listen to different English,

different accents, people who mumble.

When you study English, try to experience the diversity

by listening to a variety of different English speakers.

Another way to do this is to join online communities

and no, I'm not talking about online English language

communities, I'm talking about communities

around your hobbies and special interests

but where all the members are speaking in English.

Now this strategy is awesome for advanced

and ambitious English students and what I love about it

is you can personalise your English practice

and make it about something that you're

really interested in.

You'll learn vocabulary and expressions

that are really relevant to the conversations

you're going to have.

Since we're focusing on common natural expressions

in this lesson,

I want to show you a short video clip

of a conversation I had with my friend, Sim.

Sim!

Emma! It's so good to see you!

Fancy seeing you here!

Well I live here now.

What do you mean?

I thought you were living in Melbourne?

We just got a job and I've just moved in.

Really?

Yeah!

Do you need a hand?

Well most of our stuff's already been moved in but

would you have a car that I could borrow?

Yeah I definitely do. We've got two actually.

I live in number twenty-three

so just come grab the keys anytime.

I'll come down and see you.

It's so good to see you!

So good to see you too!

You heard a few different expressions there.

Now I want to take you a little deeper

into some of them

so that you know when to use them

and how to say them.

Let's go back to the very first thing that I said

when I saw Sim.

This expression is a common one but it has

two different meanings

depending on the tone used when it's spoken.

So the first meaning is a genuine expression of surprise

when you meet someone that you know

in an unexpected place. It's like

'What are you doing here?

I'm so surprised to see you! I wasn't expecting it at all.'

Now the second meaning is a sarcastic one.

So it's a sarcastic greeting when you see someone

somewhere where it's really not surprising at all.

Okay? It's not surprising to see them there.

So here's an example.

Every Friday, no joke, every Friday,

I go to this delicious bakery near my house

to get a phenomenally good pastry and a coffee.

Now I only allow myself to go there on Fridays because

it's so good and it's so close

that if I don't have rules I would go there every day.

So Friday is the day I'm allowed to go.

And most people know that about me,

all of my neighbours know, all my friends know

so if they ever run into me on a Friday morning

at the bakery, it really won't be a surprise.

They might say:

Well, fancy seeing you here.

Like it's not a surprise at all. I'm there every Friday

but my expression was different during my conversation

with Sim. I was genuinely surprised to see her, right?

In a positive way. I didn't expect to see her there

and you can tell by the tone. As women, we tend to

raise the tone of our voice when we get excited, right?

Very excited to see each other.

Did you notice that all of the words were stressed

when I said them?

We don't really hear any of those sounds reduce

down, we hear everything quite clearly.

In the multi-syllable words you hear one stressed

syllable that's louder and clearer than the other one.

The other one reduces down.

Now you might hear that /ng/ sound reduced down

to become the /n/ sound when it's spoken, right?

It's a little easier to say, right? So it happens

quite a bit in spoken English.

Hear how the pitch stays high?

Now in the sarcastic version, it sounds a little different.

It would sound like

'Well fancy seeing you here.'

So hear that tone changing? The meaning is different.

I'm saying that I'm not actually surprised at all

to see them in that place. The pitch is lower.

You'll probably need to add an eye roll there

just for maximum effect, right?

Let's look closer at another expression I used

during that conversation.

What do you mean?

I thought you were living in Melbourne?

I'm using this expression because I'm surprised

and confused.

At the time, I thought that Sim lived in Melbourne

so it was strange

to see her at my apartment building, right?

What are you doing here?

I'm surprised, right?

'What do you mean?' is a great expression to show

that you're a little confused.

It suggests that you need a little more information

to understand what's happening, what's going on.

Any kind of confusion can be cleared up

by asking this question.

What do you mean you're busy?

See? In all of those situations this phrase

is super helpful to clear away any confusion, right?

Now if you watched this lesson here,

another lesson about common expressions,

then you've already practised the pronunciation of

these three words with me.

Pronounced naturally, it sounds more like

The unstressed vowel's reduced down

and the words link together.

You might also hear these words expressed as

or

Right? Depends on the English accent.

Now, 'mean' is the most important word here

in this question so it's stressed.

We hear it really clearly

but all of the others reduce down.

It also helps if you put a confused face on as well.

So once I found out that Sim had only

just moved into the building,

I wanted to offer my help, right?

Like a good friend would.

Moving house is hard work right? Have you ever done it?

In my life, I've done it over twenty-five times

so of course, I know how arduous that task is

and like a good friend, I offered to help out.

It's a little more formal or more polite to say:

But Sim's an old friend of mine so I'm speaking

informally.

So this idiom is a really common way of

offering to help someone,

offering to give someone a hand is a nice way of saying

or

It's a nice thing to say if you hear someone

saying that they've got a lot of work to do

or they're stressed out or they're

struggling to do something.

And you can also use it to casually ask for help

yourself by making it a question.

Now just like in the last question,

we have the auxiliary verb 'do' and the pronoun 'you'.

Both are unstressed so when they reduce,

it sounds like

Or again you might hear that sound.

When I ask that question, Sim says that

most of their stuff had already been moved in, right?

Well, most of our stuff's already been moved in but

She's politely letting me know that she

doesn't need my help

but if she did want to accept my offer, she could say

can follow an offer for help.

Now, technically we already know that the person

doesn't mind because they offered in the first place right

But this is a nice, polite way to accept their help.

You know how painfully polite

we can be in English, right?

Right? Very polite. You can also use 'Would you mind?'

to ask someone for their help

before they've offered. Right? You can say

or

That would be so nice of you!

Notice that the main verb following this expression

is always using the -ing form, not the to-infinitive form.

Right? It's incorrect to say

Instead, make sure you're using 'subscribing'

Would you mind subscribing to my channel?

So to sound natural, very cool and relaxed

when you use this expression, you need to link 'would'

and 'you' together. So when 'would' and 'you' are linked,

again, we hear this sound come in.

Okay? 'Would' and 'you' sounds like

Can you hear that schwa sound at the end as well?

Because 'you' is the unstressed word.,

we reduce it down to the schwa.

So now I want you to practise a little.

Try using some of the expressions

that you learned during this lesson.

Write some sentences in the comments

AMERICAN B2 SPIRIT STEALTH BOMBER NOW WITHIN STRIKING DISTANCE OF CHINA - Duration: 5:55.

Pacific Air Forces Public Affairs has revealed, the US has deployed three B-2 Spirit bombers

and 200 airmen to Joint Base Pearl Harbor-Hickam in Hawaii for training in the Pacific.

The B2s were deployed to the Hawaii from Whiteman Air Force Base and are intended to support

US Strategic Command's Bomber Task Force mission.

US Strategic Command's Bomber Task Force mission is a deterrence undertaking aimed at maintaining

regional peace and reassure allies.

Lt. Col. Joshua Dorr, the director of operations for the 393rd Bomber Squadron stated,

"Deploying to Hawaii enables us to showcase to a large American and international audience

that the B-2 is on watch 24 hours a day, seven days a week ready to protect our country and

its allies.

This training is crucial to maintaining our regional interoperability.

It affords us the opportunity to work with our allies in joint exercises and validates

our always-ready global strike capability."

The latest deployment marks the second time B-2 Spirit bombers have been deployed to Hawaii.

The deployment comes at a time when there is increased tension between U.S & China and

clearly indicates America's willingness to stand up to China.

B2 Spirit brings a whole new level of capability to the table.

Let's get started.

The B-2 Spirit was developed to take over the United States Air Force's (USAF) vital

penetration missions, able to travel deep into enemy territory to deploy their ordnance,

which could include nuclear weapons.

The B-2 is a flying wing aircraft, meaning it has no fuselage or tail.

The blending of low-observable technologies with high aerodynamic efficiency and large

payload makes B 2 Spirit the most potent bomber in the world.

Even after around two decades from its introduction on 1 January 1997, there is still nothing

to match it.

America's rivals Russia and China have so far not been able to field a proper stealth

fighter let alone a stealth bomber.

Chinese have no radar that can detect or track it.

B-2 Spirit has a maximum speed of Mach 0.95.

The U.S. Air Force reports B2's range as approximately 6,900 miles or 11,000 km.

With this range, it can reach mainland China if required.

B2 has an excellent payload, which makes even one of these bombers to be extremely destructive.

It has 2 internal bays for ordnance and payload with an official limit of 40,000 lb (18,000

kg) and maximum estimated limit of 50,000 lb (23,000 kg).

Apart from different types of conventional bombs, B 2 can carry 16× B61 or B83 nuclear

bombs on Rotary Launcher Assembly (RLA).

Immunotherapy & CAR T Cells: Part 1 - Duration: 4:43.

- Our immune system has the ability to detect

and fight infectious pathogens.

It can also detect when normal cells

become cancerous and kill those cells,

preventing cancer progression.

But over time, cancers can evolve

and evade the immune response.

Researchers have been investigating

several potential avenues to counter

tumor immunosuppresion and re-activate the immune system.

One of these is CAR-T therapy,

which is designed to harness the potent cytotoxicity

of T cells by re-engineering their surface receptors

to recognize and attack tumors.

Two CAR-T therapies for the treatment

of malignant B-cell lymphomas were approved in 2017.

More recently, solid tumors such as prostate cancer

and glioblastoma have appeared on the radar

as candidates for CAR-T immunotherapy.

This is Part One of a two-part video series.

In this video, we'll cover the rationale

and design of CAR-T cells.

Okay, let's take a look at how CAR-T cells are engineered

to understand how they can enhance anti-tumor immunity.

Although there are many variations,

the general theme behind the design of CARs

is to combine two functions in one synthetic receptor.

This is done by taking moieties,

or pieces of different proteins,

and combining them into a new recombinant protein,

the chimaeric receptor.

First, the CAR ectodomain is derived

from a recombinant single change variable fragment,

or scFV, of a monoclonal antibody

that targets a tumor-specific antigen.

This provides the first function, target recognition.

The intracellular domain is derived

from a costimulatory receptor,

or from other immune receptors.

This facilitates the second function, T cell activation.

The recognition and activation functions

are grafted together on either side

of a spacer and transmembrane domain

to create the synthetic CAR transgene.

The receptor is engineered to specifically target

surface tumor antigens,

independent of MHC antigen presentation,

which is often impaired in tumors.

Activated CAR-T cells release cytotoxic compounds

like perforin and granzyme B that kill the target cell

and facilitate tumor eradication.

Activation also facilitates other immune functions

including proliferation and production

of inflammatory cytokines.

So, how could CAR-T therapy be implemented?

One paradigm is to reprogram the patient's own T cells

to express the CAR transgene.

In this scenario, blood is collected so that T cells

can be isolated and activated in the clinic.

The CAR transgene is inserted into the genome

of these cells, using a replication-incompetent virus.

Then, T cells that stably express the CAR

are isolated and expanded before being transfused

back into the patient's bloodstream.

Ensuring that the CAR-T cells are functional

is critically important

in preclinical development and beyond.

Thus, the surface expression of the CAR

needs to be monitored, as well as markers

for T cell activation such as CD25 and CD69,

and T cell exhaustion markers, such as TIM-3, LAG3 or PD-1.

Assays to characterize CAR-T cytokine release,

proliferation, and cytotoxicity

will also need to be routinely performed

throughout the process.

For example, xenograft mouse models have been employed

in preclincal studies to assess tumor killing ability

of CAR-T cells in vivo.

this is Niles, and I normally make videos on writing and literature, so make sure

to subscribe if that sounds like your thing. Now on to today's video! This is a

tag that I found on the Authortube community Facebook page, and to be honest,

I found it hilarious. So I decided I wanted to do it as well. Okay so here's

how it works. I have chosen 30 fictional characters and put their names in this

lovely fruit bowl here that I now just realized is far too big for this purpose.

The characters are chosen from the following books movies and TV shows just

to get some variation.

*Niles growls* Swedish: Nej.

We will go through 15 questions and for each of these

we will pick two names out of the bowl and I'll have to choose between

them. It's fairly straightforward. So let's get started! Question number one

Oooh, this is exciting! Option number one: Vanessa Ives. Option

number two: Jason Stackhouse.

I'm not really sure, I think Vanessa Ives is

probably better at spelling. Question number two.

... or mr. Darcy!

I wouldn't want to kill any of these!

Those were terrible options. Which one would I kill first I have a better

chance of surviving? I suppose mr. Darcy because he's a bit of a dandy and I

Today we're going to work on the pronunciation of final M

To produce the M sound

we close our lips and release the air through the nose - M

Like the sound we do when we are meditating

M is not a problem at the beginning of the words

but it might be a problem at the end because some people might not pronounce it and

just nasalize the vowel before it

We nasalize a sound when the air comes through our nose. In Portuguese, we do that a lot

for example, pão

mãe and then we transfer this to English and instead of some we say sã

And remember, there are no

nasal vowels in English

hello this is Carrie Jordan from the Rise Collective and today I want to talk to

you a little bit about how you can view goal-setting as spiritual at the end of

the video I have a process I'm going to share with you that can really help you

dive into your desire before you set your goals that you can be clear on what

you're really yearning for okay so let's get started what I've been noticing is

that some soulful spiritual creative women that I speak to altogether skip

the process of goal-setting and writing down their goals and it seems like in

many of these communities there is a belief that achievement is not is

somehow not aligned with spirituality or femininity and one one thing that I hear

from a lot of women is I want to be in flow with the universe and I want to

tell you i 100% believe in being in flow with the universe allowing room for

magic to unfold and at the same time I do not support being complacent in your

life that's the distinction being in flow at the universe and goal-setting

are not equally exclusive there's a third way it's possible to set goals and

be in flow with the universe this is where I'm going to give you the process

for a feeling into your your desires and working with your goals so first when

you're working with your desires anytime you're working with your desires or your

goals when you're diving deep into them make sure that you're coming from a

mindset of abundance and here's a tangible way that you can do that write

down a list of 20 desires that you have 20 things that you yearn for and here's

the catch make every other item something that you already have so this

is helping you to have gratitude for what you already have and come from a

place of abundance second step is feel your desire and welcome the desire and

realize that you don't always have to follow it

this is the key you just have to feel it here's a tangible way you can do that

right down your desires unfiltered just free right on a bunch of pages of paper

and then go through what you wrote and circle the ones that you're serious

about following and get focused on those one two three desires and then make

those into goals or habits now that you've written down all your desires

you've honed in on a few that you want to focus on and you're going to set your

goals don't think about how it's going to happen think more of the specifics of

what why and when of the goals and write all of these down so that you can see it

every day and make it tangible create your action plan backwards by harvesting

wisdom from yourself now I've made a little meditation for you to work with

The DJI Smart Controller helps you to get up in the air quicker and more conveniently than ever before.

When using DJI Smart Controller for the first time,

take the control sticks and screw them in,

then press and hold the Power button to turn it on.

The DJI Smart Controller has two extra function buttons compared to the Mavic 2's remote controller.

Return button (Function button),

Press once to return to the previous page,

and press twice to go back to the home page.

Long press to view the Combination button function menu.

Confirm button (Customizable button),

Press once to confirm the selection.

Before the flight, ensure that the drone has been connected to the DJI Smart Controller.

swipe down on the touch screen to show the shortcut menu,

Tap "Linking" to make the controller enter linking status.

Alternatively, you can use the button combination method,

press down the C1, C2, and Record button to enter the linking mode.

Then press the Linking button on the drone and wait for the Mavic 2 to automatically link with the controller.

After linking, launch the built-in DJI GO 4 app to control the drone.

The DJI Smart Controller's control method is the same as other remote controllers.

Push the control sticks to the inner corners to start the motors,

then gently push the left throttle stick upward to take off.

To land the drone move the left throttle stick downward for about 3 seconds.

When using the DJI Smart Controller in a bright environment,

turn on SRE mode to easily increase the screen's brightness.

Or you can press and hold the Function button on the left,

DIY Mermaid Wine Bottle Craft | Ep.2 Painting & Highlighting - Duration: 10:53.

Hi, my crafty fam!

Welcome Back to DIY No Need To Cry with Ivelisse

DIY Mermaid Wine Bottle Craft, Ep.2 Painting & Highlighting

Hi, my crafty fam!

This is where we left off in episode one.

In this episode, I'll show you how to do three different variations of colors and highlights.

I'm super excited at how these are turning out and can't wait to show you what they

look like with color.

So let's get started!

Before we start adding paint, I wanted to add more detail to the fin.

This part is optional, you can skip it if you want.

Using a pen, I drew out the pattern I wanted.

Taking some black puffy fabric paint, I traced each line.

I used a cotton swab to straighten out the lines.

Once you've outlined the entire pattern, you can create the spiky bits at the end of

the fin.

Let that dry and do the same thing to the other side.

I continued to add more height to the spiky bits until I was happy.

It should look something like this when you're done.

I decided to use black paint to prime the whole thing this time instead of the usual

white because I thought the color shift paints would look better on the black.

I'm not really if it made a difference or not.

Paint the whole bottle including the fin and let it completely dry.

Now using three different colors and three separate pouncer brushes to paint, start painting

the tail.

I started with the pink in the middle

Then I used the purple on the outside as you see here

did the same thing to the other side

brought the purple down a little

Then the green

and back to the pink.

from here on I just kept alternating the colors until I reached the bottom.

Let it dry and give it another layer.

With this layer, and any layer after, you want to make sure to be neat with the paint.

I gave my bottle about three coats, letting dry between each one.

Before adding the highlight color, I gave it a coat of polycrylic to seal it.

You really want to seal it before adding the highlight if you have a light color such as

the pink.

I'll show you why in the next step.

Once it was dry I added the silver highlight to this bottle.

I started by adding some to the fin very lightly.

You want to rub it into the creases of each scale.

You want to do this, in one motion, going against the scale, so that it all piles into

the crease.

I used a damp cloth to wipe the excess off my fingers.

This is why you had to seal it before adding the highlight… because it will ruin the

pink if you don't.

The sealer gives it a slippery coat so it doesn't stick to the pink… too much...

and if it does… you're able to clean off any of the excess paint.

I added silver glitter glue the same way I did the highlight and let that dry.

When I was done doing that I added the chunky glitter to the tail.

Lastly, I highlight the entire edge of the fin with the silver

and here she is all painted, well it… not quite a mermaid yet now is she lol

I'm loving how these are coming out so far but I have to say that the silver was the

hardest of all three highlights.

This is not the original bottle I showed you in the live stream.

I ended up messing that one up when I added the highlight because I didn't seal it first

and it ruined the pink.

This is the gold highlight with gold glitter and different shades of paint

and this is the pearl highlighted one with chunky iridescent glitter.

I did them all exactly the same I just used different color combos.

So there you have it my crafty fam, three different color combos you can paint these

and as usual, you can paint it however you'd like.

be just because it's a new app that you could put on your phone most of the

people have had any kind of issues here they're always getting responses from

the app makers telling them what to do and how to fix it and you know if

something's been misinterpreted or they haven't don't think they've gotten paid

or they think it's a scam or whatever the cases I'm seeing here just like you

can see here TV - it explains to you in detail like you know what the problem is

what you need to do where the menus are and the same thing with down here they

did the same thing down here someone said is the scam they didn't get paid in

36 hours and then this person basically gets an explanation as to why or where

you can go on your phone to find out what your status is so these people

aren't afraid they're not running they're not hiding they really want this

to work and they're answering basically all the

bad questions or negative questions or comments that they're getting in under

this app in Google Play Store as you can see look it was just updated on the

fourteenth and today's the 15th as I'm recording this now it has over 5,000

installs okay the size is 22 megabytes which is really small program and it's

really easy and simple to use and one thing I do want to point out to anyone

who's seriously into this like in the Bitcoin or cryptocurrency okay if you

have any questions at all besides you're lying and any kind of feedback you may

got on here on this app check out these websites okay especially I would say

Facebook and Twitter because people leave comments on Facebook and Twitter

especially if they're negative or positive and it's good for you to read

through those and get a really good feel and a really good idea of whether or not

this actually this app is something that it's good for you now from what I've

read and from what I've gathered from looking at this you need to earn 50

Speak Life Into Your Endeavours - Duration: 5:32.

Are you speaking life or death into your endeavours?

Everyday we speak into existence, things we desire, or that which we do not care for,

it may not specifically mean that if we say "Wealth Appear Now" it will happen, but, if

over a period of time you, not only speak about an appearance of wealth in your life;

however, you GENUINELY believe and, most importantly, perform positive actions which may help lead

your life towards a path of wealth, then it is possible, you will see yourself physically

headed into that direction.

As the saying states "Energy Flows Where Attention Goes".

In the past, when relationships failed to work out for my cousin, instead of moping

herself to smithereens with sorrow, she would find opportunities to uplift herself.

Sometimes it would involve her keeping really busy with work, studies and other valuable

items in her life.

These are choices she made, as she recognised the value she IS.

This is the bottomline.

When we recognise the valuable individuals we are, we see how important our contributions

are to the collective.

With this awareness, it makes things easier for us to overstand the importance of injecting

life into our endeavours, but does this mean we will not experience difficulty?

No, I will be frank, this practise does NOT guarantee we will not have challenges.

What it DOES guarantee is our reaction to these challenges, which may HELP us instead

of hurt us, as we get ahead.

Well, what is this whole "speak life into endeavours all about"?

Here are a few examples: 1.

The "Poor Me" scenario.

This one is very common, "poor me this didn't happen", "poor me that didn't happen", "oh

my gosh, you mean to tel me ... (fill in the blank) is happening to me again?

Poor me".

More than crying negativity onto oneself, the lack of ability to make a breakthrough

is prevalent.

One has already flooded oneself with 'poor me's', also, the energy one has built around

being so sorry for oneself makes it near-impossible to even recognise opportunities standing around

awaiting selection.

The "Broke Song".

"Oh guuuuud, I cya pay for dat nah, I rel brokesss" full stop.

Not only is admitting how broke one is, can be a way of giving a temporary situation permanent

housing, but we are reminded to pay attention to our thoughts, as they become words, which

become actions.

Which so ever actions one wishes to implement in one's life, it is important to speak it

into reality, as soon enough, one's actions begin mimicking one's belief.

The "I'm Not Good Enough" Gang.

Hmm, once guilty, once charged - No More.

In our local setting, we often believe we are not worthy of superb things.

Often times when we speak of doing the unthinkable immediately, we revert to inferiority.

This may be possible because of the amount of work necessary to achieve such, or, we

think due to living in smaaall Caribbean islands we are not worthy of doing grandddd things.

I know for sure the size of our islands and our populations are not limitations of the

great things we are capable of.

Caribbean people make the list of The World's Finest often.

I know the phrase "if you believe you will achieve" can easily sound like a broken record,

but in reality, as we apply ourselves, we do conquer great things.

Another point is that often, we become too concerned with other people's opinions and

therefore remain limited, because we do not desire others, thinking too much of us, as

some of us unfortunately do not believe we are much ourselves.

The journey of greatness entails many closed doors, failures, restarts and challenges,

but do you know what comes from these closed doors, failures, restarts and challenges?

Greater opportunities, exceptionally valuable lessons, growth AND success.

Experiencing failure easily makes us timid amongst others, but if we do not fail during

our efforts of building grand things, we miss opportunities of gaining vital experiences,

while overcoming mediocrity.

Seeking superb growth requires making grand gestures, which may place us in temporary

discomforts, this proves to be worthwhile over time, once we are dedicated.

As we speak life into our endeavours, we do so with refreshed mindsets, contributing greatness,

as we have originally been equipped with this gift from birth.

> Let's go ahead and talk about those because there are a number of symptoms

that we need to look at, and the first one is that heavier prolonged menstrual

bleeding that comes in there but let's go ahead and look at the graphic and if

you could give us an ID here of what we're seeing. So, so basically when we say

heavier prolonged mass or bleeding it's that that goes beyond the eight days,

that becomes more frequent during the month, that you actually are bleeding.

Some of my patients come to me and tell me look I'm having a period twice a

month that is not normal, even if it was normal for you for the last year or so

it is not normal. That needs to be evaluated this pain pressure the

fullness in the pelvis again this is likening to that of fibroids that are

big enough will feel like you're carrying a baby, and for those who

haven't carry baby will feel like there's pressure in that area where

there wasn't before. Frequent urination or constipation, these fibroids when

they're large enough are pressing on your bladder as I spoke a little bit

before, can even make you have some incontinence and if they also press on

the bowel and that really in the bladder and the bowel they cause problems of

incontinence and pressure. During intercourse, this is a large uterus it's

behind it, so basically they're in intercourse there's pressure on on

intercourse and pain. That should not be normal and should warrant an evaluation,

and then if you can feel your abdomen to be enlarged you can feel that there's a

mass absolutely that that needs to be evaluated. > Can you find the fibroids in a

normal pelvic exam? find them all? > No and that's... even for us gynecologist that we

do this many times a day. Sometimes the fibroids as we saw on that graphic

before can be inside the uterus, can cause a lot of bleeding and pain, but you

still have a normal-sized uterus. You should if they're large enough feel that

there's a problem as a gynecologist and refer your patient for a pelvic

ultrasound.

Hello everyone, and welcome to my channel. If you are new here my name is Arnella

this is Niles, and I normally make videos on writing and literature, so make sure

to subscribe if that sounds like your thing. Now on to today's video! This is a

tag that I found on the Authortube community Facebook page, and to be honest,

I found it hilarious. So I decided I wanted to do it as well. Okay so here's

how it works. I have chosen 30 fictional characters and put their names in this

lovely fruit bowl here that I now just realized is far too big for this purpose.

The characters are chosen from the following books movies and TV shows just

to get some variation.

*Niles growls* Swedish: Nej.

We will go through 15 questions and for each of these

we will pick two names out of the bowl and I'll have to choose between

them. It's fairly straightforward. So let's get started! Question number one

Oooh, this is exciting! Option number one: Vanessa Ives. Option

number two: Jason Stackhouse.

I'm not really sure, I think Vanessa Ives is

probably better at spelling. Question number two.

First one is ...

Mrs. Weasley

... or mr. Darcy!

I wouldn't want to kill any of these!

Those were terrible options. Which one would I kill first I have a better

chance of surviving? I suppose mr. Darcy because he's a bit of a dandy and I

would be truly terrified if Mrs. Weasley came at me to be honest.

What a shame that Mr. Darcy is already spent.

Wow, okay, so Gandalf comes with magic and

Lord Grantham comes with the Downton Abbey estate. Which one would I give my rose to?

Mr. Collins is a minister so I would say Mr. Collins, even though he would probably

bore me to death, but still, I don't think Cersei Lannister will comfort me.

Or Melisandre.

Definitely Melisandre, I feel like she would know how to form a group around

herself, for sure!

Ooh, let's see.

Hmm, none of these two really feel like they would be very good at remembering

birthdays. Black Widow would just be off with the Avengers fighting bad guys

whereas Gollum would just be thinking about the ring all the time... and yet I'll

still say Gollum.

Whereas Dorian Gray - Dorian Gray's a bit more into the luxurious side of life so I think

can't figure it out.

The Warriors Look Mortal, But What Does That Mean? - Duration: 8:18.

 Thursday in Oakland was about as good as the NBA gets. Behind James Harden (44 points, 15 assists, 10 assists) and Clint Capela (29 points, 21 rebounds) and well-timed cameos from Danuel House (17 points), Austin Rivers (18 points), and Gerald Green (16 points), the Rockets came back from a 17-point halftime deficit to push the Warriors to overtime on the road

Then, possession after possession, Houston answered every single Golden State bucket in overtime

Along the way there were apparent Steph daggers, egregious missed calls, and Bird Box tweets

It all ended when Harden hit a step-back three over the outstretched arms of Draymond Green and Klay Thompson to give Houston the lead with one second left

 The whole game had turned into its own little 1 a.m. fever dream by the final minutes, but that Harden shot was particularly mind-blowing

The Rockets have now won of 11 of their past 12 games and Harden is averaging 40

1 PPG over that stretch; he's on another planet right now. "Let's make it clear," SI's Rob Mahoney wrote earlier this week

"Harden is one of the greatest offensive players to ever play the sport." The past month in Houston has left almost no room for debate

 As for the Warriors, though . There's plenty of room to wonder. Golden State was on its heels for much of the second half Thursday, and as the game got close, the offense reverted to the same iso-heavy muck that we watched during last year's Western Conference finals

After a first half that seemed to make a mockery of anyone who thought Golden State had real problems, the rest of game was the kind of disjointed loss that's become common over the past few months

AP Photo/Ben Margot  Curry, Durant, and Klay Thompson combined for 87 points Thursday, but Draymond Green, Andre Iguodala, and Shaun Livingston were largely ineffective on offense

Golden State got almost nothing from the rest of the bench. Curry (35 points) got only one shot over the final 3:48 of the fourth quarter, while Durant went 2 of 7

Golden State fans on Twitter were once again aghast at Steve Kerr's rotations and his continued refusal to put KD and Steph in the pick-and-roll

In general, it was all weirdly familiar. This season's Warriors seem to have more problems than years past and fewer easy answers

Maybe this is just how the regular season works for them now.  "We're maybe the most scrutinized team in the history of the league," Steve Kerr said last week

Leaving aside the inevitable fact-checking that claim invites, it's fair to concede that Kerr is making a valid point

Year after year, the current version of the Warriors have inspired concern trolling from fans and media all over the world

I'm sure some of that gets old, and there are dozens of "What's wrong with the Warriors?" discussions that have come to look ridiculous by the end of the past few seasons

So let's not do that.  If the question is what these Warriors struggles mean for this season, the answer is probably nothing

Klay Thompson has been struggling, but he should be fine. His defense remains outstanding, and he's shooting 51% from the field and 58% from three over the past three games

That's a small sample size, but it's also a reminder that betting against Thompson's jumpshot is a horrible idea

 SHAPIRO: Draymond Green Hasn't Been Himself on Offense  Draymond, meanwhile, is now a shooter that the whole league is betting against on a regular basis

He's been left wide open all year, and he can't knock down shots. That one is a real issue

But the caveat with Draymond is that he's still an all-world defender and a flat-out maniac who should deliver in the playoffs

Last season Green was on cruise control for six months during the regular season, and then he spent May and June leading the Warriors in minutes, rebounds, assists, blocks, and steals

Next to a healthy Durant and Curry, Draymond's shooting isn't fatal; he'll help set the tone when it matters

 And speaking of Durant and Curry, their partnership has always been a little bit more awkward than it should be, but it's always been close to unstoppable when titles are on the line

These two are the new-age Kobe and Shaq, but with two additional stars running beside them

If KD and Curry are healthy, Golden State probably isn't losing. (Also: DeMarcus Cousins is coming back soon

Nobody knows what he'll look like on the court, but if he's anywhere close to what he was, this could get unfair quickly

Don't be one of those "Will DeMarcus Cousins make the Warriors worse?" people.)  The much better Warriors discussion is what happens next season

There is league-wide speculation that Durant will leave after this year in Golden State

Two months ago, many around the league would have answered those rumors by wondering whether the Warriors would still be title contenders even without KD

But if there's anything that's changed this season, or if there's anything we've really learned, it's that the post-KD future in Golden State isn't nearly as secure as many assumed

Remove Durant next year, and this is a team whose best role players (Iguodala, Livingston) are getting old and less effective

 WOO: NBA Draft Big Board 3.0: Ja Morant Rises Up the Ranks   The bench has become a little thinner every year

Boogie is likely gone after this season, and there won't be very much room to make meaningful improvements around Klay, Curry, and Draymond

And then, yes, Draymond: it's one thing to downplay his struggles when he's the fourth star on a team that has three of the best shooters of all time

If he becomes the third superstar on a team that has limited room to add firepower anywhere else, the equation becomes more complicated

The entire team would be relying on Curry to do more than he ever has, with less help than Golden State has given him since 2013

 All of this would open the door for a slew of imperfect challengers that have been easy to dismiss over the past few years

I'm thinking of teams like the Thunder, Bucks, Nuggets, Raptors, Celtics, Lakers, or Rockets; depending on where Anthony Davis ends up, the league's almost-superteams may actually have a chance next season

We'd all have to recalibrate how we grade contenders, because the impossible Golden State curve won't exist anymore

 Until then, of course, Cousins returns soon, Klay is already rediscovering his jumper, Durant and Curry are two of the four best players in basketball, and Draymond is still one of the best defenders in the world

This spring the Warriors will likely run through the playoffs and once again make the whole league look helpless

 Mortality may be looming, but let's not get ahead of ourselves.
